Abstract
The invention is applicable to the technical field of biological analysis, and provides a monoclonal antibody IgM de novo sequencing method based on mass spectrum, which comprises the following steps of providing a monoclonal antibody IgM sample; subjecting the sample to reduction and alkylation; the method comprises the steps of carrying out enzymolysis on a treated sample by using specific endoprotease, carrying out analysis by adopting a liquid chromatography-mass spectrometry combined technology to obtain mass spectrum data, carrying out de novo analysis on the mass spectrum data by utilizing a data processing algorithm to deduce the amino acid sequence of the monoclonal antibody IgM, obviously improving the quality and stability of a peptide fragment product through an optimized sample pretreatment flow, obtaining more comprehensive and accurate mass spectrum data by adopting a high-resolution mass spectrum platform and an innovative SIM-DIA acquisition mode, particularly improving the detection rate of a low-abundance peptide fragment, and realizing high-efficiency and accurate analysis on complex mass spectrum data by adopting data analysis software and an algorithm which are independently researched and developed, thereby greatly improving the efficiency and success rate of de novo sequencing.
